Lonza’s group of structural protein scientists will provide you with the ideal combination of in silico tools and expert knowledge for optimal antibody humanization, deimmunisation and complete protein engineering. In combination with our industry leading expression systems (mammalian and microbial), Lonza supports early stage customers in their protein molecule optimization and testing as well as proof of concept material needs. This provides customers the reassurance of obtaining and working with representative material by using the same expression systems throughout discovery and development to GMP manufacture.





Antibody Humanization and Protein Design:  

Antibody Humanization

Starting from antibody sequences derived from non-human origins, antibody structure models are built using homologous search and in-house antibody structure database. A panel of variants is generated to humanize the antibody whilst respecting antibody structural integrity and avoiding potentially risky post-translational modifications (PTMs). In combination with our proprietary Epibase™ T cell epitope screening tool, candidates can be designed to further minimize the potential immunogenicity risks. A small quantity of material can be generated and tested for binding affinity and soluble aggregate as well as using in vitro assays to assess the potential of unwanted T cell responses (optional) to compare candidates and obtain the best leads.

  • Antibody structure modeling
  • Identification of high-risk, post-translational modifications and chemical stability motifs
  • Humanization and deimmunisation of full-length and antibody fragments
  • Expression and characterization of humanized (and deimmunised) variants
    • For antibody fragments, suitable expression host system can be tested using Rapid Host Selection Services
  • In vitro T cell testing of variants using human PBMCs (optional)


Protein Design

Using various proprietary and public in silico modeling tools, we build protein and antibody models, screen for undesirable features, identify desirable substitutions that modify and enhance your protein's properties such as its activity, affinity, specificity and stability. In conjunction with Lonza's Epibase™ platform, immunogenic hot spots can be identified and removed to obtain an optimal product with a good safety profile.
